【求助】帮忙诊断脑BrdU非特异性染色的问题

可能灌流的时候没弄好（猜测出现自溶），有些切片如下图1,2有很多非特异的染色，不知还有什么方法可以补救。

染色的protocol

0.9%生理盐水灌流，4%PFA固定
0.1 M PB 3*5min
2N HCL in 0.1 M PB at 60°C for 30min
0.1 M PB 5*5min
0.6% H₂O₂ and 10% 甲醇in 0.1M PB for 10 minutes
0.5% Triton-X 100 and 10% Normal Goat Serum in 0.1 M PB for 1 hour.
1:5,000 rat anti-BrdU antibody and in a 0.5% Triton-X and 2% NGS in 0.1 M PB at 4 °C for two nights.
incubated for 2 hours at room temperature.
5*5 min in 0.5% Triton-X in 0.1 M PB
1:300 dilution of biotin-conjugated goat anti-rat antibody in 0.5% Triton-X 100 and 2% NGS in 0.1 M PB for 2 hours.
5*5 min 0.1 M PB
ABC kit for 90 minutes.
