NEW YORK (Reuters Health) May 23 - Caspase-14 protects the skin against ultraviolet B photodamage and water loss, according to the results of studies in caspase-14 knockout mice, researchers in Belgium and the US report.

Drs. Declercq and Vandenabeele and colleagues investigated the role of caspase-14 by generating caspase-14-deficient mice by homologous recombination. Although caspase-14 expression was ablated in all epidermal tissues, there were no gross histological abnormalities in the deficient mice.

Deficiency in caspase-14 impaired the cleavage of (pro)filaggrin to filaggrin, which has an important role in epidermal barrier function, the researchers note. As a consequence, transepidermal water loss was about 20% higher in caspase-14-deficient mice.

"The observed differences between normal and caspase-14-deficient mice underscore the fact that caspase-14 is an important player in the skin protection against the DNA damaging properties of ultraviolet light and water loss," Drs. Declercq and Vandenabeele conclude.

"Also during skin transplantation, application of caspase-14 or modulation of its expression by pharmacological molecules could accelerate the skin barrier recovery."
