多发性骨髓瘤（MM）具有肿瘤和骨病兼并的特点，严重危害着患者的健康与生命。Wnt/β-catenin通路参与MM的肿瘤增殖和骨质破坏过程，最新研究表明骨硬化蛋白（Sclerostin）能抑制Wnt/β-catenin通路，是MM靶向治疗研究的新热点。我们前期研究发现：毒结清口服液可兼治MM肿瘤和骨病，且能下调骨硬化蛋白。据此我们推测Sclerostin串联Wnt/β-catenin通路，可能是毒结清口服液兼治MM肿瘤和骨病的作用机制。本研究拟以人MM细胞株分别构建细胞和小鼠模型，以人MM细胞株凋亡及荷瘤小鼠骨髓间充质干细胞向成骨细胞转化为观察靶标，采用不同剂量毒结清口服液干预，观察其对MM骨硬化蛋白(包括特异性激活和抑制）和Wnt/β-catenin通路的影响以及兼治MM肿瘤和骨病的效果。研究有望阐明毒结清口服液对MM肿瘤与骨病标本兼治的新机制，为MM的中医临床治疗提供技术支撑。

multiple myeloma (MM) ，as a life-threatening malignancy, has the clinical features that including both the cancer and bone disease. The canonical Wnt/β-catenin pathway plays an important role in the process of proliferation and bone destruction in MM.The latest research shows that sclerostin inhibit Wnt/beta -catenin pathway, which has been a new target in  therapy of MM. Our previous study found that Dujieqing Lliquor can be used for treatment of  tumor and myeloma bone disease, and can reduce the bone sclerosis protein. Therefore, it is suggested that the Wnt/ beta -catenin pathway in combamination with bone sclerosis protein may be the mechanism of treating MM tumor and bone disease by Dujieqing Lliquor. Our study intends to construct human MM cell line  and mouse model with apoptosis in human MM cells and mice bone marrow mesenchymal stem cells into osteoblasts as target, using different doses of oral drug settlement intervention, to observe the effect of expression of sclerostin (Including specific activation and inhibition) and affiliated protein of Wnt/ beta-catenin pathway. The study is expected to clarify the new mechanism of MM and to provide technical support for the TCM specimens and treatment of MM.
