【求助】Th1/Th2平衡

想研究基因A对Th1/Th2平衡的影响，由于之前没做过T细胞这方面的研究，而且免疫学的也不太好，不了解T细胞的主流研究方法，设计预实验很大阻力，求战友帮忙指点啊。
我现在想到的方法：
1. 取PBMC然后过表达和敲减A检测T-bet和GATA3的变化（PBMC鱼龙混杂，感觉说明不了问题）
2. Th1细胞过表达和敲减A后检测T-bet、IFN-g的变化
3. Th2细胞过表达和敲减A后检测GATA3、IL-4、5、10、13的变化
CD4+纯真T细胞来源：人脐带血？
Th1细胞来源：CD4+纯真T细胞通过IL-12诱导分化
Th2细胞来源：CD4+纯真T细胞通过IL-4诱导分化
有篇文献的protocol是这么写的，大家觉得主流麽？
Ex Vivo Polarization of TH1 and TH2 Cells
CD4+T cells were cultured in RPMI 1640 media supplemented with 10% FCS (Invitrogen). Cells were cultured in 24 well plates precoated with 0.5 mg/ml of anti-CD3 and in the presence of 0.5 mg/ml soluble anti-CD28 (eBiosciences). TH1 polarization was induced using 2.5 ng/ml IL-12 and TH2 polarization with 10 ng/ml IL-4 (R&D Systems). After 48 h of polarization, 17 ng/ml of IL-2 (R&D Systems) was added. Polarized cells were grown further for various time points from 24 to 72 h.
是否有必要用诱导分化成熟的Th1和Th2细胞做实验？（诱导分化感觉挺麻烦的。）有没有可供研究的相关细胞系啊？
