Disodium Cromoglycate Active Against Respiratory Syncytial Virus
NEW YORK (Reuters Health) Mar 06 - Disodium cromoglycate, an agent commonly used to prevent bronchial asthma and allergic rhinitis, reduced the symptoms of induced respiratory syncytial virus (RSV) infection in mice, in a study by Japanese researchers reported in the February issue of the Journal of Medical Virology. The results suggest that disodium cromoglycate (DSCG) could be an effective agent for preventing RSV-induced disease and/or relieving the symptoms of RSV infection in humans.
A member of the Pneumovirus genus of the family Paramyxoviridae, RSV causes respiratory-tract infections in patients of all ages and is the leading cause of respiratory failure in young children. Natural infection with RSV does not confer immunity.
The report notes that earlier studies of DSCG's effects on viral infection had been inconsistent. Some research showed that DSCG is a useful adjunct therapy in alleviating asthma symptoms and in preventing colds in adults with asthma, while other research suggested that it is not effective against viral infection either in vitro or in vivo.
Dr. Koichi Hashimoto of Fukushima Medical University and fellow researchers divided a cohort of 8-week-old female BALB/c mice into four groups: RSV-infected and then treated with DSCG; RSV-infected and then treated with saline; mock-infected with RSV and treated with DSCG; and mock-infected with RSV and treated with saline. The groups treated with DSCG received 3, 10 or 30 mg/kg/dose/day.
The mice that received the lower two doses of DSCG were significantly protected from RSV-induced weight loss, compared with the mice that received only sterile saline (p < 0.05).
The mice that received the highest (30 mg/kg/day) dose of DSCG were also protected against weight loss significantly more than those that received 3 mg/kg/day (p < 0.05).
In addition, the study found that although mice treated with DSCG at the time of the RSV inoculation had a decreased lung viral titer on day 1 post-infection versus saline-treated mice, this difference was not maintained.
Based on these findings, along with those from an in vitro portion of the study, the researchers suggest that DSCG inhibits RSV replication in vitro, but acts against RSV by a different mechanism in vivo, possibly through immunomodulatory and anti-inflammatory effects.
