Postdoc fellow-stem cell aging and leukemia
A postdoctoral fellow position is available at the University of Kentucky School of Medicine Department of Toxicology and Cancer Biology,to investigate stem cell inflammaging and leukemia in blood system. The research and cancerous conditions. Usinggenetic mapping approach,we identified the first quantitative trait gene, latexin,whose natural variation negatively correlates with HSC number variation in the population(Nature Genetics,2007).We further uncovered several novel fuctions and underlying molecular mechanism underlying the regulating of latexin in normal,stress and malignant hematopoiesis.
(i)We discovered the potential tumor suppressor fuctionof latexin in leukemia and lymphoma(Plos One, 2012).
(ii)We identified ribosomal protein subunit 3(Rps3) as a novel latexin binding protein. Latexin increases Rps3 nuclear translation and sensitize cancer cells to rediation(Cell Death&Disease,2014).
(iii)We uncovered that latexin deletion in vivo increased HSC survival and refeneration by downregulation of Thbs1 signaling(Stem Cell Reports, 2017).
(iv)We also first the first time found that latexin transcription is regulated by both cis- and trans- factors. A single nucleotide polymorphism(SNP) is directly associated with its differential expression in HSCs. HMGB2 acts as a trans-factor and negatively regulates latexin transcription(Haemato;ogica, 2019).
Our current efforts of latexin-related research are focused on two aspects:
(i)screening of small molecule compounds that activate and inhibit latexin activity;
(ii)understanding the extrinsic role of latexin in bone marrow niche in regulating normal and malignant hematopiesis.
The other research focus of my laboratory is hematopoietic and immune aging. We found that old HSCs have deceased homing effciency(Blood, 2005 ), and old HSCs drive ageing-related immune modeling(Blodd, 2018)Both genetic and epigenetc mechanisms may contribute to the aging of stem cells in hematopoietic system, which may underlie the organism aging and longevity(Stem Cell Translational Medicine, 2012). The current research interests are focusd on aging and infection-induced cytokine storm,particular alarmins, and their effect on hematopoietic and immune aging.
Candidates should have a Ph.D or equivalent degree and experience with investigating biochemistry, cell biology, molecular biology, as well as mouse models.

目前,我们的研究主要集中在两个方面: (i)筛选激活和抑制该基因活性的小分子化合物
ii)了解该基因在骨髓生态位调控正常和恶性造血中的外在作用。
候选人应具有博士学位或同等学历,并具有研究生物化学、细胞生物学、分子生物学以及小鼠模型的经验。如有细胞测序分析经验也可。入职后我们将提供具有竞争力的薪酬,博后及以上预计($55,000-$75,000 );与资历稳定性和学校规定有关。
